# What gets quieter as we get older?

We look at four age-related changes in the body, keeping the measured signal separate from the much bigger question of whether replacing it helps.

## The same word can hide different measurements.

“Lower” sounds simple until we ask lower where, compared with whom and measured how. Blood and muscle are different places. A concentration is different from a hormone released in pulses. An average across age groups is different from a measurement that follows the same person over time.

These observations do not establish a common rate of decline or an age at which treatment becomes necessary.

## NAD+: a helper inside cells

NAD+ belongs to the coenzymes, helper molecules that assist enzymes; it is not a peptide. Enzymes are proteins that help chemical reactions happen; NAD+ takes part in energy chemistry and other cellular processes. The research describes [age-associated decreases in tissue NAD+](https://pubmed.ncbi.nlm.nih.gov/24360282/), with cell and mouse experiments investigating possible mechanisms.

A tissue measurement cannot by itself establish what an infusion does inside that tissue. Nor does evidence about an oral precursor, a starting material the body can convert, automatically describe NAD+ administered directly. The [NAD+ guide](/nad-plus/) separates those routes and questions.

## MOTS-c: blood and muscle tell different stories

MOTS-c is a peptide whose genetic instructions come from mitochondria, the structures involved in cells’ energy supply. The body makes it, and researchers have measured it in blood and muscle. In [a study of healthy aging men](https://pubmed.ncbi.nlm.nih.gov/32182209/), circulating levels were lower in the older group while muscle levels were higher.

That detail matters. “MOTS-c falls with age” would leave out half the observation. The study did not give MOTS-c to people to see whether a symptom changed. It cannot establish a need for treatment, even if the natural signal deserves further research. Our [MOTS-c page](/mots-c/) keeps the animal experiments separate from the human measurements.

## Growth hormone: a pulse is not a reservoir

Growth hormone is released in bursts. GHRH, short for growth hormone-releasing hormone, is one of the signals that prompts the pituitary gland beneath the brain to release it. [Research in younger and older men](https://pubmed.ncbi.nlm.nih.gov/10522985/) links aging with less nighttime growth-hormone output and suggests a contribution from reduced GHRH signaling.

Sermorelin copies a portion of GHRH. It is neither growth hormone itself nor a replacement for every change in the system. A smaller pulse does not establish an adult disease. The [sermorelin guide](/sermorelin/) explains why its former approval for children does not answer the adult aging question.

## GHK: the measured peptide and the copper complex

GHK is a chain of three protein building blocks. GHK-Cu is GHK holding copper. A [review of GHK and aging](https://pmc.ncbi.nlm.nih.gov/articles/PMC8789089/) describes lower plasma GHK with age, while discussing cell and animal research. The measurement concerns GHK; the proposed preparation may be GHK-Cu. The distinction is part of the question, not a typographical nicety.

There is no established inference from less plasma GHK to a benefit from a GHK-Cu prescription. Our [GHK-Cu reading](/ghk-cu/) considers the evidence without borrowing promises from cosmetic marketing.

## An observation does not choose a prescription.

Promise Peptides provides a U.S. route for questions about NAD+, MOTS-c, sermorelin and GHK-Cu. A licensed provider reviews each request rather than treating an age bracket as a prescription. Compounded preparations are not FDA-approved, and the review may find the person ineligible.

We can acknowledge a biological pattern without accepting the sales argument sometimes attached to it. What remains unknown includes who, if anyone, gains a meaningful long-term result from the matching option, which risks accumulate and what comparison would answer those questions fairly. Age gives the question context. It does not supply the answer.

## A question can be the next step.

Promise Peptides connects U.S. patients with licensed U.S. providers who examine every request and decide whether prescribing is appropriate. The review may find that a person does not qualify. Access differs with the patient’s state and the treatment. Licensed U.S. compounding pharmacies prepare prescriptions; compounded medicines prepared for these prescriptions are not FDA-approved.
